_Second in charge of the laboratory.? _Checking and issuing of test results.? _Application of the restriction fragment length? polymorphism (RFLP) technique to HLA-DR typing for routine laboratory
use. The same technique was also applied to HLA-DPB, -DQ, -DX, and -DOB
typing.? _Introduction of polymerase chain reaction (PCR) based techniques such
as sequence specific oligonucleotide (SSO) typing, and amplified fragment
length polymorphism? (AFLP) typing for the determination of HLA-DRB1, -DRB3, -DRB4, -DRB5,
-DP, -DQB1, and DQA1 typing.? _Monitoring of the DR and DQ subtyping methods to maintain the highest
degree of subtyping possible. This also involves the development of methods
to apply sequence? specific priming (SSP) as a problem solving technique in HLA class
II subtyping.? _Programming of an algorithm for the determination of HLA alleles from
restriction enzyme results.? _Programming of a Laboratory data base to store typing results and
issue reports.?

_The success obtained in DNA class II typing has resulted in the routine
use of these techniques in matching for organ transplantation.? _Participation in the collaborative transplant study (CTS) organised
by Professor G. Opelz (Germany) which has lead to several publications.? _A Population study involving DNA-DR and -DQ typing of?Papua
New Guineans and Australian Aborigines (published).? _Development of HLA-DRB5 typing (published) and HLA-DRB3 typing using
the AFLP technique.? _Discovery of new sequences in the HLA-DR and -DQ region (submitted).? _Further studies are being carried out examining the HLA class II genes
in various Asian and South East Asian populations which will be of benefit to patients
from these ethnic groups who require transplantation.?#060;/TD>
